Abstract

Systemic sclerosis (SSc) is a rheumatic autoimmune disease of unknown etiology, characterized mainly by vasculopathy, autoimmunity and fibrosis of the skin and internal organs. The accumulation of collagen fibers is the most prominent feature in SSc pathogenesis, with the increase in the expression of type V collagen (Col V), already in the early stages of the disease. In recent years, Col V has been associated with autoimmunity in several diseases, when exposed to the immune system in tissue remodeling processes. In SSc an important participation of this protein in the pathogenesis of the disease is suggested, since an anomalous Col V deposition in the skin and lung of SSc patients was shown, related to the disease activity. In addition, the immunization of healthy rabbits with Col V to develop an experimental model similar to SSc, with histological and immunological characteristics similar to the disease in humans, which are reversed after induction of nasal tolerance with Col V. This study aims to develop biochemical and enzymatic cleavage of Col V, to study the immunogenicity of this protein in SSc. The Col V fragments will be characterized according to their molecular weight. The antigenic potential of Col V in SSc will be evaluated by immunoblot using rabbit sera from the SSc model before and after induction of nasal tolerance with Col V. Fragments with higher antigenic potential will also be identified by immunoblot, with 6 sera from patients with anti-Col V antibodies. (AU)
