Photodinamic inactivation using curcumin conjugated to Pluronic ®F-127 on Streptococcus mutans and Candida albicans biofilm

Abstract

The photodynamic therapy is described as a promising treatment to inactivate oral microorganisms. Therefore, curcumin is a hydrophobic photosensitizer that can compromise the efficiency, the development of studies that investigate the effect of curcumin when associated with polymeric micelles are important. Thus, the purpose of this study is to evaluate the efficacy of photodynamic inactivation (PDI) on a Streptococcus mutans and Candida albicans biofilm using curcumin conjugated or not with micelles as photosensitizer, irradiated by a LED light source. The fluorescence and optical absorption spectroscopies, the minimal inhibitory concentration (MIC), minimal bactericidal concentration (MBC), and minimal fungicidal concentration (MFC) will be determined. Single species biofilms will be induced using a 96-well plate followed by duo species biofilms. Colony forming unit (UFC/mL) will be obtained after determination of MIC, MBC and MFC. The interaction and cellular viability of the biofilms will be analyzed by confocal microscopy using BacLight® LIVE/DEAD dye. Different Groups will be evaluated: PSM+L+ (Photosensitizer + micelles + light), PSD+L+ (Photosensitizer + DMSO + light), PS-L- (negative control), PS-L+ (Light group), chlorhexidine 2% (positive control) and nistatine (positive control). After evaluation of normality and homoscedasticity, if they are met, the data will be submitted to analysis of Variance (ANOVA) and multiple comparison, otherwise, non-parametric analysis will be used.
