Influence of murine IgG repertoire on the maturation of B and gamma-deltaT cells with modulatory and/or regulatory potential in vitro

Abstract

Over the last 15 years our research group has been studying the effects of maternal immunization with allergens. At this time, several mechanisms has been suggested to elucidate the influence of maternal immunization, in this context direct interactions between maternal IgG and offspring lymphocytes during its development in primary lymphoid organs has been highlighted. Preliminary results of our group suggested B and ³dT cells are more susceptible in vivo effects of maternal immunization. Based on these evidences, this project aim to elucidate this mechanism in a in vitro condition. Therefore, purified IgG produced in response to diverse antigens will be used to evaluate its effect upon thymus and bone marrow cells. By means of flow cytometry, we intend to evaluate phenotypic and intracellular cytokine production alterations in different maturation stages of the investigated populations. Using these approaches, we intend to elucidate the potential of IgG exerting a pivotal role upon B and ³dT cells maturation. Taken together, the results obtained from this project can collaborate to the future development of strategies on the immune system regulation. (AU)
