Abstract

The filamentous fungus Trichoderma harzianum has great biotechnological importance, being considered as a biocontrol agent, besides the ability to hydrolyze lignocellulose components efficiently. Preliminary studies have proven the hydrolytic ability of strain CBMAI-0179 and the high enzymatic activity related to the degradation of cellulose and hemicellulose of aqueous extracts. Transcriptional analysis of Trichoderma harzianum CBMAI-0179 during cellulose degradation will provide important information about the genes involved in the depolymerization of cellulose chains promoted by the fungus. These informations can be compared with other strains and other species of filamentous fungi with the potential of biomass biodegradation, which should help to establish genes and proteins that are important for biodegradation reactions. This project proposes to conduct a deeper study of the gene set related to carbohydrate degradation, classified according to the Carbohydrate-Active Enzymes Database - CAZymes, induced by the fungus growth using cellulose as carbon source, comparatively with the gene set from glucose growth. Functional annotation, the study of the expressed genes and the ratio of the expressed genes to the detected proteins from the extract will be performed. Thus, it is expected to identify the main mechanisms of gene regulation responsible for the different enzymatic activities observed. (AU)
