Abstract

The definition of a heterogeneous population of spermatozoa within the ejaculate of different species of mammals has been intensely studied in the last decade. The identification and quantification of sperm subpopulations has been shown as an important evaluation to predict the fertilizer capacity of the animal and to explain problems of subfertility of some animals. However, no study was found to correlate the sperm subpopulations with the in vitro production of embryos in bulls. With the widespread growth of the recently introduced Senepol breed in our country and the scarcity of studies evaluating the seminal quality of bulls of this breed, this research project will be carried out with the objective of evaluating the relationship between the sperm subpopulations in the cryopreserved semen of Senepol bulls with the in vitro production of embryos. Forty commercial doses of semen from Senepol bulls, selected on the basis of retrospective data from in vitro embryo production, will undergo a Percoll gradient and will be evaluated by Computer-Assisted Sperm Analysis (CASA) to define the kinetic parameter of each sperm and, from this analysis, the existing sperm subpopulations will be identified and quantified. In addition, the integrity of the plasma membrane will be performed using fluorescent probes and sperm morphology using the wet preparation technique. Retrospective data on the in vitro production of embryos will be obtained from the records belonging to the company Senepol Mais and In Vitro Brasil. Linear and nonlinear models already described in the literature will be tested to establish the relationships between the sperm subpopulations defined by sperm kinetics and embryo production. The variables obtained in the evaluation of membrane integrity and sperm morphology will be added to statistical techniques of partial least squares regression, linear discriminant analysis and artificial intelligence, seeking a better accuracy in the prediction of the production of embryos of the bulls.
