Abstract

The widespread use of synthetic fragrances in industrialized domestic products, together with the lack of studies related to airway poisoning, constitute a scenario in which it becomes indispensable to carry out studies capable of establishing effective methods for identifying possibly toxic substances present in those products. This work will evaluate, through in vitro tests, from human bronchial epithelial cell culture (BEAS-2B), the toxic effect of synthetic fragrance, such as cinnamyl alcohol, commonly found in cleaning products, cosmetics and perfumes, whose exposure through the air may be responsible for triggering a number of adverse health effects. The aim of this research is to develop a methodology for the toxicological study of synthetic fragrances, verifying the role of detoxifying enzymes in the cytochrome P450 family (CYP1A1, CYP1B1, CYP1A2, CYP2A6 and CYP2A6 and - 2A13) responsible for the metabolism of fragrances in BEAS-2B culture samples. The effect of cytotoxicity will be assessed by MTT test and lactate dehydrogenase (LDH) assay. It will also be performed, by means of extraction of total RNA and the reverse transcription (RT) of the cells, quantitative real-time PCR to determine the expression of genes of the CYP450 system.
