Abstract

Cd concentrations in agricultural soils in the humid tropics can represent a serious socio-environmental problem, since Cd is toxic for most plants, animals and humans. However, it is possible to reduce the Cd concentration in the soils by phytoremediation, especially if we understand how the Cd bioavailability in the soils can be influenced by the cultivation of phytoremediation plants of this metal in different climatic seasons. Thus, our objective with this study will be to evaluate the Cd bioavailability in an Oxisol cultivated with Brachiaria decumbens Stapf. cv. Basilisk and Panicum maximum Jacq. cv. Massai, in different climatic seasons (summer and winter conditions). The relationships between the Cd bioavailability and the tolerance mechanisms of these plants that favor the Cd accumulation will be evaluated, in order to optimize the process of Cd phytoremediation, by using Noccaea caerulescens (J. & C. Presl) F.K. Mey as a model plant. Plants of Brachiaria decumbens cv. Basilisk, Panicum maximum cv. Massai and Noccaea caerulescens will be cultivated in an Oxisol contaminated with Cd (3.6 mg kg-1 soil) during a single growing period in each climatic seasons. Then, the following procedures will be carried out in soil samples: (i) physicochemical and geochemical analyzes (in synchrotron light), (ii) determinations of Cd concentrations and Cd fractionation in the soil, (iii) calculations of factors related to Cd phytoextraction, (iv) determinations of the contents of nutrients and Cd in plants (including Cd in apoplast and symplast in the roots), (v) measurement of root length and root surface, (vi) measurement of reduced glutathione (GSH) and phytochelatins (PCs) concentrations, (vii) measurement of malondialdehyde concentration and of the activity of the enzymes superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (GPX) and glutathione reductase (GR), and (viii) measurement of photosynthetic variables.
