Arctium lappa glycolic extract: antimicrobial activity against anaerobic bacteria of interest to dentistry

Abstract

The antimicrobial action of numerous herbal can bring benefits to dental therapy, however, there are still very few researches that analyze the effects of plant extracts on anaerobic microorganisms involved in oral infections. The objective of this study was to evaluate the antimicrobial action of the bardana glycolic extract (Arctium lappa) on the anaerobic bacteria Porphyromonas gingivalis and Fusobacterium nucleatum. Initially the Minimum Inhibitory Concentrations (MIC) and Microbicide (CMM) of the bardana extract for the bacterial species evaluated will be determined, according to Clinical and Laboratory Standards Institute (CLSI). Subsequently, the action of the effective concentration of this extract will be evaluated on monotypic biofilms of these bacteria formed in microtiter plate wells for 7 days (37ºC in anaerobic chamber). The biofilms will be exposed to two different concentrations of the extract for 5 min, 30 min and 24 h, with n = 10 for each experimental group. To control will be used saline solution 0.9% (C +) and solution of clorexidine 0.12% (C-), totalizing 4 groups for each bacterium analyzed, in each experimental time. After this the biofilms will be disaggregated by sonication (25% of potency / 30 s), the suspension generated will be serially diluted (1:10) and seeded in Brucella agar. After 48 to 72 h (37oC in anaerobic chamber) the concentration of CFU / mL (forming units per milliliter) will be determined. The data obtained will be analyzed statistically by ANOVA and Tukey (pd0.05).
