Abstract

The emergence of antibiotic resistance is considered a threat to human and animal health. Through horizontal trasfer, environmental antibiotic contamination may promote the exchange of genes between non-pathogenic and pathogenic environmental bacteria. The resulting remodeling of existing microbiomas leads to evolutionary and environmental complications that in association with their dissemination abilities classify genes of antibiotic resistance (ARGs) as environmental contaminants. Seabirds rapidly respond to environmental changes, being excellent marine ecossystem health biomarkers. Wildlife rehabilitation centers have a controverse role on the possible dissemination of bacteria presenting pathogenic genes and ARGs, both in the internal environment and external environment, after these animals are released. We intend on evaluating the degree of human interference in the marine ecossystem through the presence, identification and subsequent quantification of seleced ARGs (tetA, tetB, tetY, tetK, tetM, tetQ, sulI, sulII, str, aadA, catI, catII, erm(B), erm(F), qnrB, qnrS, blaCTX, blaTEM, mecA and mcr-1) present in plasmids of the microbiome of free ranging wild seabirds of two distinct groups: admitted in rehabilitation centers of the southern brazilian coast and residents of seabird colonies in the southeastern coast. For this purpose, we will employ real-time PCR (rtPCR) on fresh fecal samples (enema) collected (I) upon the individual's admission to the rehabilitation center; (II) 24 hours after initiation of the antimicrobial treatment; (III) 24 hrs after antimicrobial treatment termination and (IV) upon release, and of E. coli colonies previously isolated from several seabird species and stored in the Laboratory of Wildlife Comparative Pathology - LAPCOM/FMVZ/USP. Our goal is to determine the degree of environmental contamination by the ARGs present in seabirds at their habitat/admission, as well as the influence of antimicrobial treatment and/or stay in rehabilitation centers in regards to the presence and quantity of ARGs.
