Abstract

Tissue regeneration by using biodegradable macro-porous scaffolds is considered a new treatment approach on Medicine and Dentistry. The application of this therapy aims the stimulation of local mesenchymal stem cells (MSCs) differentiantion, leading to neo-tissue-genesis. Therefore, the present study aims to develop scaffolds with different porosity degree and evaluate the adhesion, spread and proliferation of MSCs seeded onto its surface. The scaffolds will be prepared by pH modulation of chitosan solution, followed by freeze at different temperatures, and freeze-drying. The materials architecture and porosity will be evaluated on images obtained by scanning electronic microscopy and analysis on image J software. After that, the MSCs will be seeded tonto the scaffolds to evaluate adhesion and spread, by F-actin immunofluorescence, and cell proliferation, by DNA quantitation. Numerical data will be subjected to statistical analysis.
