In vivo effect of Low-Intensity Pulsed Ultrasound (LIPUS)in bone neoformation of calvaria defects in ovariectomized rats

Abstract

Osteoporosis is characterized by a decrease in bone mineral density, resulting in reduced force and trabecular bone weakness. Drug therapy to avoid bone loss in these cases is commonly used, despite the possibility of side effects issues. Thus, the development of non invasive strategies based on biophysical stimulation to better bone regeneration is been recently investigated. The purpose of this study is to perform in vivo assays to evaluate the stimulatory potential of low-intensity pulsed ultrasound (LIPUS) in calvaria defects created in osteoporotic rats. There will be utilized 36 wistar female rats divided in 4 groups (n=8): C (control), CLIPUS (control + LIPUS), OVX (ovariectomized) and OVXLIPUS (ovariectomized + LIPUS). After 90 days of ovariectomy, there will be created unilateral bone defects of 5mm diameter at the left side of parietal lobe of all animals. During the following 4 weeks, the selected groups will receive LIPUS application (1,58 MHz, 200¼s pulsation, 1KHz frequency and intensity of 40 m/w/cm2 for 20 minutes three days a week on two selected points on skin defects. After 30 days, the animals will be euthanized to collect calvaria samples to perform qualitative and quantitative histologic analysis. The obtained data will be analyzed by adequate statistical tests with the significance set to p d 0.05, with the goal to find positive effects with the utilization of LIPUS.
