Abstract

Adult human brain organotypic cultures are a powerful method for studying cellular and molecular aspects of neuropathologies, as well as the effects of neuroprotective drugs. The main advantages of this method include maintaining the brain structure and its cellular connectivity. However, there is still a lack of studies that better characterize this model, not only to obtain more reliable results, but also to expand its applications. The objective this present project is to expand the characterization of a model of adult human brain using cortical slices obtained from patients undergoing surgery for epilepsy, observing the oxidative metabolism of this tissue by respirometry and citrate synthase activity. We will also assess the maintenance of neuronal function over the days in culture by assessing c-fos protein levels. Furthermore, we will apply our model in studies of infection of the central nervous system by Oropouche virus, determining infected neural phenotypes, apoptotic markers and possible cellular parameters that can be altered as a result this infection.
