Abstract

UV radiation is associated with cutaneous phototoxicity, early photoaging, photoimmuniosuppression, and carcinogenesis in an increasingly prevalent way. Although photoprotective products have proven SPF (Sun Protection Factor), their effectiveness and safety also rely on formulation composition and how it does interact with the skin. With increasing complexity of sunscreen formulations as well as UVA and UVB radiation exposure, concerns are raised over the underlying mechanisms of photoprotective activity and its risks. Avobenzone is one of the few consolidated organic UVA filters with limitations on photo-stability and safety. It is also commonly associated with octocrylene photo-stabilizer, and with inorganic filters, such as titanium dioxide. In this context, the present study aims to develop and to comparatively evaluate formulations of organic and inorganic UV filters, isolated and associated, containing avobenzone, octocrylene and titanium dioxide UV filters as active substances. Formulations will be submitted to evaluation of the photoprotective clinical efficacy from UVA and UVB radiations, by determining SPF in vitro and in vivo, and of the effects over structural and morphological characteristics of epidermis and dermis, by confocal reflectance microscopy and high frequency ultrasound, and of UV filters penetration by skin image analysis using confocal reflectance microscopy and a Raman confocal spectroscopy.
