Abstract

The main objective of the present study is to evaluate the application of low temperature plasma at atmospheric pressure (LTAPP) for the control of cariogenic biofilms. To this end, the effective physical parameters of LTAPP against polymicrobial biofilms composed of combinations of the species Streptococcus mutans, Streptococcus gordonii, Streptococcus sanguinis, Lactobacillus casei, Candida albicans and Actinomyces viscosus will be established. Multi-species biofilms will be submitted to LTAPP and quantitative and microscopic analyzes (confocal, scanning electron microscopy and fluorescence) will be performed. The action of LTAPP on biofilms formed in situ with different degrees of cariogenicity will also be evaluated. Experimentally induced caries cavities will be submitted to LTAPP, restored and evaluated. Negative control (no treatment) and positive control (chlorhexidine 2%) will be included. The cellular effects of LTAPP on S. mutans, L. casei and A. viscosus will be studied by scanning electron microscopy and Fourier Transform Infrared Spectroscopy (FT-IR). Finally, the cytotoxicity of LTAPP at effective conditions on gingival fibroblasts and primary culture of fibroblasts from the dental pulp will be evaluated. All assays will be performed in triplicate in three independent experiments. The results will be tabulated and analyzed for distribution. The most appropriate statistical tests will then be selected.
