Abstract

Microorganisms and their by-products are proved to be responsible for the development of periapical pathologies whether in primary or in secondary/persistent endodontic infections. It is important to understand the pathogenicity mechanisms between these root canal bacteria and host in the presence of apical periodontitis. Furthermore it is mandatory the comprehension of the relationship between root canal content and the periapical region in cases of absence of endodontic infection and heath periapical conditions. Thus, the aim of this study is to characterize by Mass Spectrometry the proteome of 30 root canals of teeth with different endodontic situation, as: a) vital pulp; b) primary infection and c) secondary/persistent infection. Ten patients with periapical heath and indication of endodontic treatment for prosthetic reasons, 10 patients with apical periodontitis and primary infection and 10 patients with apical periodontitis and need for endodontic retreatment were selected. Samples from the root canal content were collected and will be further processed by Capillary Nano-Flow 2-Dimensional Liquid Chromatography and Electrospray Ionization Tandem Mass Spectrometry (2D-LC-ESI-MS/MS). All the proposed work involving mass spectrometric analyses will be performed using the in-house LTQ-Velos-ESI-MS/MS instrumentation (Thermo-Scientific, San Jose, CA). The acquired MS/MS spectra generated will be separately searched against specific bacterial and human protein database (Swiss PROT and TREMBL, Swiss Institute of Bioinformatics, Geneva, Switzerland, http://ca.expasy.org) for all samples using Proteome Discoverer 1.3 software and SEQUEST algorithm. The data will be tabulated and statistically analyzed.
