Polysaccharides and special metabolites from basidiomycetes fungi: isolation, characterization and evaluation of antitumor potential

The study of basidiomycetes, mushrooms also known as macroscopic fungi, as a biosprospective resource of medicinal products, has developed the interest of a large amount of researches across the world. This interest is due to the production of a large amount of bioactive substances, from primary and secondary metabolism. These substances from fungi metabolism has been extracted and studied due to its antitumor potential through evaluation of cytotoxic potential, and consequently, evaluation of its immunomodulatory potential. Among these compounds obtained from primary metabolism of fungi, polysaccharides, specially ²-glucans, is described by its immunomodulatory potential as they attach to cell receptors of the immune system, as well as to induce in some defense cells the synthesis of compounds, such as nitric oxide. However, secondary metabolites of basidiomycetes as terpenes, alkaloids, steroids and benzoic derivatives, also have antitumor potential. Therefore, this project suggests the isolation and identification of metabolites, through extraction, chromatographic fractionation and structural characterization of the compounds by using spectrometric and spectroscopic techniques. After being isolated and identified, compounds from primary and secondary metabolism will be submitted to evaluation of its antitumor potential against different tumor cell lines, in order to detect cytotoxicity, as well as genic expression assays to detect immunomodulatory potential of the isolated polysaccharides. Additionally, cytotoxic assays will be carried out to evaluate the cytotoxic potential of secondary metabolites against MCF-7, HT-144, A549, HepG2 and HTH 83 tumor cell lines. (AU)
