Multicentric clinicopathological and proteomic study of adenomatoid odontogenic tumors

Abstract

Odontogenic tumors are uncommon in the practice of oral pathology and have clinical behavior that varies according to their clinical, radiographic and microscopic characteristics. Adenomatoid odontogenic tumors (AOT) are expansive, slow-growing and progressive tumors, located commonly in the anterior region of the maxilla of children. Ameloblastomas are odontogenic tumors with potentially infiltrative and aggressive biological behavior, with considerable rates of recurrence, located mainly in the posterior region of the mandible of young adults, demonstrating mutations in BRAFV600E and SMO genes. Ameloblastic carcinoma (AC) represents the malignant counterpart of ameloblastoma and a third of patients develop long-term lung metastases. Proteomics analysis has been used to reveal the large-scale protein expression of various neoplasms, however, this technique has not yet been applied in the context of odontogenic tumors. The aim of this study is to evaluate the clinicopathological and proteomic profile of AOT, ameloblastoma, and AC of three countries, identifying prognostic markers for aggressive lesions. Twenty samples with representative material will be collected from paraffin blocks acquired from the archives of four Oral Pathology laboratories in Brazil, Guatemala, and South Africa, with 10 cases of AOT, 5 cases of ameloblastoma and 5 cases of AC that will be submitted to mass spectrometry linked to liquid chromatography for the proteomic study. Protein identification will be based on the UniProt library. Bioinformatics analysis to determine the sub- or super-expression of each component will be carried out using different software, such as the MaxQuant package and SIMCA 14.1. The prognostic potential of the proteins that obtained a statistically significant result will be validated by means of immunohistochemical reactions in a convenience sample composed of twice the number of cases of each tumor, through correlation with clinicopathological data and survival. The use of these techniques may contribute to the treatment of affected patients. (AU)
