Abstract

Prostate cancer (PCa) is the second most frequent and with a higher rate of morbidity and mortality among men. Studies highlight the role of intestinal metaplasia in the process of tumor progression and invasion. However, more studies are needed to understand how this metaplasia is related to the development of castration-resistant PCa. Thus, this project aims to describe the incidence, multiplicity, and characteristic of androgen independence of intestinal metaplasia in prostate cancer of transgenic mice for the Pten gene. Histological sections of prostates from wild type and transgenic animals will be performed at different ages and will be stained by Hematoxylin and Eosin and by the Periodic Acid-Schiff method. In addition, immunohistochemistry for the androgen receptor and the ki-67 proliferation antigen will be performed. Incidence and multiplicity will be determined by counting the numbers of foci of intestinal metaplasia per animal and prostate lobe. The index of proliferation and androgen receptor-positive cells will be also determined in the foci of metaplasia and in the adjacent areas. These results will contribute to a better understanding of the participation of intestinal metaplasia and revealing if it may be related to the process of tumor progression and the onset of castration-resistant prostate cancer.
