Abstract

Deprivation of ovarian hormones in young rats is marked by an important increase in cardiac fibrosis, accompanied by adverse cardiac remodeling. However, the mechanisms are still uncertain. Thus, we will investigate the involvement of the renin-angiotensin-aldosterone system (RAAS) in these findings, and whether the mechanisms involved are linked to increased activity and expression of matrix metalloproteinase-2 (MMP-2) and calpain-1. In addition, we will also investigate the role of aerobic physical training as moderator of this process. Sixty-four Wistar rats were submitted to ovariectomy at the 10th week of life and were divided into two groups: a group of 22-week-old sedentary rats (N= 32) and a group of trained rats (N= 2) during the last 10 weeks (12th to 22nd week of life). Each large group will be subdivided into two smaller groups; treated with vehicle (N =16) and treated with enalapril maleate (10mg/kg/day, N=16), an angiotensin II converting enzyme inhibitor. All animals will be submitted to the following experimental protocols; hemodynamic evaluation will consist of recording the evolution of blood pressure (BP) and heart rate (HR) by means of plethysmographic examination at four different times, 12th, 14th, 18th and 22nd weeks of life; echocardiographic examination at the 12th and 22nd weeks of life to assess cardiac morphology and functionality; evaluation of the activities of calpain-1 and MMP-2 using gel zymography, and protein expression by means of western blot and immunofluorescence techniques.
