Abstract

Breast cancer is currently the most common neoplasm in women worldwide, but there are few alternatives for its prevention, a factor that must be prioritized for disease control. In this project, we propose the development of topical nanogels for the self-administration of the combination of celecoxib and endoxifene, in order to optimize deep skin penetration and localization of drugs in breast tissue as a new strategy for the chemoprevention of breast cancer. Lamellar phase nanogels and nanoemulsions will be developed using a mixture of surfactants composed of soy phosphatidylcholine and glycerol, and the oil phase will be comprised of tricaprilin, monocaprilin and Shea or passion fruit butter. The relationship between composition, type phase formed and incorporation of the drug will be evaluated. Subsequently, selected formulations will be characterized, and evaluated for to their ability to promote penetration into the skin. Assays using the HET-CAM model will be used to estimate the potential irritation of the formulation on the skin, while the cytotoxic effect of the drugs incorporated into the nanocarrier will be evaluated in MCF-7 cells using the MTT method.
