Abstract

In Brazil and many other countries pre eclampsia (PE) stands out as the main hypertensive disorder in pregnancy, being potentially fatal for both mother and embryo and affecting 3-8% of all pregnancies in the world. However, little is known about the etiology of this pathology. It is believed that utero-placental ischemia causes the release of placental factors in the maternal circulation, which end up causing damage to the endothelium of these pregnant women through alterations in gene expression, alteration un the production of vasoactive factors and damage by oxidative stress. SIRT1 is a class III protein of histone deacetylases that exhibit NAD + dependent function. Being highly expressed in endothelial cells, this protein is one of the main molecules to combat oxidative stress in the organism, besides regulating gene expression and the release of nitric oxide (NO) by eNOS. In order to evaluate the relationship between PE and SIRT1 in vitro, human umbilical vein endothelial cells (HUVECs) will be incubated with plasma of healthy pregnant women, hypertensive pregnant women and pre-eclamptic pregnant women in the presence or absence of Resveratrol, a potent natural antioxidant and activator of SIRT1, and EX-527, an inhibitor of SIRT1 activity. Therefore, the expression and activity of SIRT1, besides its possible effects on endothelial damage, reactive oxygen species (ROS) and nitric oxide (NO), will be analyzed.
