Ultrastructural study of the equine endometrium exposed to ozone

Abstract

The present study will describe, for the first time, the uterine ultrastructural changes due to the infusion of ozone (O3) in healthy mares. A total of 21 females will be assigned into one of three groups according to the treatment (n = 7 mares/group): O3, ½O3 and Control groups will receive intrauterine treatment with O3-O3 gas mixture containing 42¼g, 21¼g and 0¼g O3/mL, respectively. The treatments will be performed every three days and during six days. In each treatment, a flow of 0.25 mL gas/min will be infused during three consecutive minutes. Endometrial fragments will be collected immediately before the first treatment and 24 hours after the third insufflation by uterine biopsy. Fixation of the endometrial tissue will be done using 2.5% glutaraldehyde in 0.1M cacodylate. Posteriorly, the uterine architecture will be evaluated by scanning electron microscope considering nine parameters: a) the structure of the luminal epithelium, b) quantification of different cell types, c) size and forms of the different cell types, d) decidualization of the stroma, e) pinopodes visualization, f) density of microvillus, g) endometrial vascularization, h ) frequency and size of grooves, and i) presence of cellular or tissue fragmentation.
