Evaluation of the inflammatory response in peripheral blood mononuclear cells of sedentary individuals: role of physical exercise and capsaicin

Abstract

Physical inactivity associated with poor eating habits is associated with excessive accumulation of visceral adipose tissue and chronic low-grade inflammation. Adoption of non-pharmacological strategies that potentiate the energy expenditure that leads to reduction of adipose tissue stores, especially visceral fat deposition and reduction in pro-inflammatory cytokine concentrations. We hypothesized that the adoption of the high-intensity interval training (HIIT) associated with the daily intake of the bioactive compounds of pepper popularly known as capsaicin (capsiate) promote protective effects through the reduction of body fat and modulation of inflammatory response of peripheral blood mononuclear cells. The objective of the present study will be to evaluate the effect of HIIT associated with capsaicin supplementation on body composition and inflammatory response of peripheral blood mononuclear cells (PBMC). The experimental design of the study will be double-blind placebo controlled. Participants will perform 12 weeks of HIIT, 3x per week. A maximum incremental test will be performed on the treadmill to quantify the maximum oxygen consumption (VO2max), body evaluation will be by DEXA and visceral fat by ultrasonography. Blood samples collected at rest for biochemical and cytokine analyzes. The PBMCs will be isolated and stimulated in vitro LPS and Concacavalin A, in the presence or absence of capsaicin and capsiate, evaluating the production of cytokines in the culture medium, and gene expression of inflammatory proteins in the PBMC after 48h of treatment in the different training periods physicist. For statistical treatment, a two-way Shapiro-Wilk test and analysis of variance will be performed to compare moments (pre and post intervention), analyzed by SPSS software (version 22.0). (AU)
