Abstract

During aging changes occur in the heart tissue supporting the development of cardiovascular disease (CVD). At the cellular level, telomere shortening and activation of some cell cycle inhibitory molecules such as p16, p21 and p53 are described as biomarkers of senescent cells. Studying the cell senescence process in experimental rodent models is challenging due to the relatively long time for chronological/ natural aging. Therefore, it is necessary to establish models that anticipate the senescence process. Recent studies have demonstrated that treatment with D-galactose is an interesting tool to induce aging in rodents. In this project we will establish a protocol for the study of cardiac tissue senescence in C57BL/6 mice treated with D-galactose, compared with a natural aging model. We will evaluate senescence biomarkers such as the expression of p16, p21 and p53, as well as possible changes in telomere length among groups. The use of experimental models that contribute to the understanding of mechanisms involved in the cardiac aging process will benefit the development of future interventions that may prevent cardiac manifestations and decrease CVD-related deaths in the aging population.
