Abstract

With the discovery of cisplatin, metals have been gaining more and more space in medicinal chemistry because they have a greater possibility of forming diverse molecules and with promising biological / pharmacological activities. In this context, silver has been a well-explored metal, as it has been used for centuries due to its antibacterial action. In view of this, the objective of this work is to evaluate the cytotoxicity and mutagenicity of the silver complex with furosemide (Ag-FSE), previously tested and with a proven antimicrobial potential to ensure safe use. The cytotoxic potential will be evaluated by the Trypan blue test and clonogenic survival using cell lines GM07492 (non-transforming fibroblastic type cell) and Caco-2 (cell line derived from colorectal adenocarcinoma) because they have lower IC50 values when evaluated by the oxide-reduction of resazurin (unpublished data - Annex 1). The mutagenic / genotoxic activity will be evaluated by the Micronucleus and Comet assays, using the HepG2 lineage, as it presents a metabolization profile. Considering that Ag-FSE is a potential candidate for the development of a new drug, this study will provide reliable data on its interaction with genetic material, through genotoxicological tests, as well as on the cytotoxic effect, ensuring the safest and most effective application for human health.
