Abstract

Head and neck cancer corresponds to tumors located in the upper aerodigestive tract. The most effective treatment is radiotherapy concomitant with chemotherapy with high doses of cisplatin; however, its use is limited due to its adverse reactions, especially nephrotoxicity. Therefore, there is a need for identification of nephrotoxicity predictor molecules, since classical pathological markers have low sensitivity and specificity. Among the proposed biomarkers in recent years, one of the most promising is miRNA. The aim of this study will be to evaluate miRNAs as possible predictors of cisplatin-induced nephrotoxicity in head and neck cancer patients. This is an analytical, experimental, clinical, single-arm, prospective and quantitative study, whose sampling was non-probabilistic consecutive. Patients with head and neck cancer who underwent antineoplastic treatment with cisplatin (80-100mg/m2 every 21 days for 3 cycles; only the 1st cycle being considered) with concurrent radiotherapy will be included. Nephrotoxicity, gastrointestinal toxicities and myelotoxicity will be assessed for severity according to CTCAE v5. The expression of plasma miRNAs in relation to cisplatin-induced nephrotoxicity will be evaluated to find a correlation that makes them predictive molecules of this toxicity. Statistical analysis will be performed using the Chiquadrado, Fisher exact, Mann-Whitney and ANOVA tests for repeated measures, considering p <0.05 as significant. (AU)
