Influence of Cisplatin-associated tamoxifen on the osseointegration of titanium implants

Abstract

The purpose of the present study will be to evaluate the osseointegration of titanium implants installed in tibias of rats modified systemically by tamoxifen (TAM) associated or not with Cisplatin (CIS). Will be used 72 rats, randomly distributed in two experimental groups (n = 36), the animals will receive bilateral ovariectomy, tibial implant installation bilaterally and after 8 weeks, administration by saline gavage (SS) and tamoxifen citrate (TAM) throughout the trial period. Twelve animals from each group will be euthanized at week 10, 14 and 18, the right and left tibias will be collected and histological, histometric, immunohistochemical and ultrastructural analyzes performed. The morphological analysis will consist of the description of the histological characteristics of the bone tissue in the threads of the titanium implant and bone/implant interface areas. Immunohistochemical markings will be evaluated semi-quantitatively for all antibodies. For histometric analysis, the following evaluation criteria will be adopted: Direct Contact (CD) of bone tissue with the implant surface in relation to the Total Length (TC) of the implant threads (mm); Neoformed Bone Area (AON) in relation to the Total Area (AT) present within the implant thread limits (mm²). Analyzes will be performed on both sides of the implant, in the cervical, middle and apical thirds. The focus of ultrastructural analysis will be the interface between bone and the implant surface. The relationship between the osteocytes and their extensions with the titanium surface and adjacent osteocytes, as well as the distribution of these cells in the periimplant bone tissue will be evaluated. Quantitative data will be submitted to parametric or nonparametric statistical analyzes with a significance level of 5%.
