Abstract Group A rotaviruses (RVA) are the main cause of acute gastroenteritis in humans. RVAs are species-specific; however interspecies transmission and rearrangement between human and animal RVAs contribute to their genetic diversity. The G8 genotype is widely distributed in animal species, being reported in pigs, horses, but mainly in cattle. The detection of G8 strains infecting humans is rare, nevertheless reported worldwide, including in Brazil. The origin of the G8 strains in the human population has been investigated in recent years and a potential bovine origin is suggested. G8 strains exhibit an ability to repeatedly combine with VP4 strains ecologically adapted to the human gut (ie, P[4], P[6] and P[8]), which could facilitate their dispersion in the human population. Human G8P[8] strains may possess both Wa-like and DS-1-like genomic constellation, and recently, G8P[8] DS-1-like strains with bovine G8 origin have emerged in Asia. The emergence of strains exhibiting unusual and/or interspecific rearrangements can lead to future implications in vaccination programs. The RVA monitoring conducted by the Adolfo Lutz Institute in the last 10 years (2009-2019) permitted the identification of 15 unusual RVA G8 strains associated with P[4] (5 samples), P[6] (7 samples) and P[8] (3 samples). The aims of the present study is to conduct the molecular characterization and phylogenetic analysis of the 11 segments (genomic constellation) of RVA G8P[4]/P[6]/P[8] strains detected between 2009 and 2017, targeting to reveal the true origin of these strains, as well as understanding their ability to evade vaccine immunity. | |
