Abstract The immersion media could influence the evaluation of different bioceramic materials properties. This study will aim to evaluate the effect of immersion media on biocompatibility, bioactivity, and physicochemical properties of bioceramic materials. Different immersion media will be used: distilled water, phosphate-buffered saline (PBS), and in vivo model in the subcutaneous tissue of rats. Bioceramic cements will be evaluated according to Subproject 1: repair bioceramic sealers (Bio-C Repair, Biodentine and IRM); Subproject 2: endodontic sealers (Bio-C Sealer, BioRoot RCS and AH Plus); and Subproject 3: new hybrid cements (repair and sealer) according to the proportion of handling - NeoMTA 2 (2 proportions), MTA Repair HP and BioRoot. Polyethylene tubes will be filled with the materials (n=7) and immediately placed in microtubes (Eppendorf, Hamburg, Germany) with different solutions. Scanning will be performed using computerized microtomography (micro-CT, SkyScan 1276, Bruker-microCT, Kontich, Belgium) to determine baseline and stored for periods of 7 and 30 days at 37ºC. The implantation of the materials in subcutaneous tissue will be performed with 28 Holtzman rats (n=7), with 4 implants per animal for subprojects 1 and 2. After the periods of 7 and 30 days, samples will be scanning again to determine volumetric change and porosity. The same samples will be analyzed by scanning electron microscopy (SEM, JSM-6610LV Scanning Electron Microscope, JEOL, Tokyo, Japan) and dispersive energy spectroscopy (EDS) to verify bioactive potential. Also, after the experimental periods, the biological effects caused by bioceramic cements will be characterized. Saos-2 lineage cells will be used which will come in contact with the cement extract after immersion in different dilutions. Therefore, the cell viability tests will be performed by the MTT and Neutral Red method, in addition to bioactivity tests by the activity of alkaline phosphatase (ALP) and red staining of alizarin (ARS). All cell analyzes will be performed in three independent experiments. For data analysis, the results will be subjected to appropriate statistical tests. (AU) | |
