Abstract

The presence of active ingredient inhibiting biofilm growth in dentifrice can provide a significant and clinically relevant effect, particularly when combined with satisfactory mechanical removal of the micriobiota. The aim of this study is to evaluate the potential for antimicrobial and cytotoxicity of dentifrices based on sodium fluoride, amine fluoride, zinc citrate, arginine and chlorhexidine. The dentifrices that will be evaluated: Colgate Periograd®, Colgate Sensitive Pro-Relief Immediate®, Curaprox Enzycal 1450®, Elmex®. For the analysis of cytotoxicity, cultures of fibroblasts from mice L929 (American Type Culture Collection) exposed to toothpaste extracts will be used, evaluating cell viability through MTT and neutral red assays. The culture medium will be used as a negative control (unexposed cells) and the 20% DMSO as a positive control. The tests will be carried out in triplicate and repeated in three independent times. For the antimicrobial analysis, an agar inhibition halo test will be performed for the following microorganisms: Streptoccocus mutans (ATCC#25175), Candida albicans (ATCC CBS-ICB/USP#562) and Staphylococcus aureus (ATCC # 25923). After the incubation period, the inhibition halos will be measured, in millimeters, three times to obtain the average for each well / disc. The area of the halos will be calculated using the formula used to estimate the area in circles (A=ÀxR2). The statistical tests will be chosen according to the distribution and homoscedasticity of the data, using a significance level of 5%.
