Effect of orchiectomy on the activities of acid and alkaline phosphatases in the salivary glands of Wistar rats

Abstract

The variation in plasma testosterone concentration emerges as an important factor in the oral health of men. Phosphatases are involved in the molecular mechanisms of various diseases, since they play a role in controlling cell proliferation, growth and differentiation. Changes in phosphatases in the salivary glands have been associated with experimental diabetes, cigarette exposure and cancer. In this context, the objective of this research will be to analyze the effects of orchiectomy on the salivary flow rate, on the salivary biochemical parameters and on the activity profile of phosphatases in the homogenates of the salivary glands of rats. For this purpose, twenty male Wistar rats (3 months old) will be divided into two groups (n = 10): SHAM, from which the testicles will be exposed, but not removed; orchiectomized group (ORX), the rats will be bilaterally orchiectomized. After 8 weeks of castration, the animals will be weighed, anesthetized to collect the saliva induced by pilocarpine and then they will be euthanized via blood exsanguination, for blood collection and later excision of the salivary glands, which will be washed with physiological solution, weighed and then store at -80 ° C until your analysis. In the collected saliva, the salivary flow rate, pH, buffering capacity, total protein concentration and amylase activity will be measured. In the glandular homogenates, the activities of total acid phosphatase, tartrate-resistant acid phosphatase, total alkaline phosphatase and total protein concentration will be analyzed. The normality and homoscedasticity of the data will be analyzed and they will be submitted to the most appropriate parametric or non-parametric test. For all tests, the level of rejection of the null hypothesis will be set at 5% (p < 0.05).
