Passive ultrasonic agitation using EDTA and it's impact at the blood clot morphology formed pos endodontics restorative procedures: in vitro study

Abstract

The aim of the present study will be to evaluate in vitro the characteristics of the blood clot in teeth submitted to different final irrigation protocols. Sixty uniradicular bovine teeth will be prepared to simulate open apexes and randomly divided into six groups according to the following final irrigation protocols: (1) Saline (SF); (2) Saline + Passive ultrasonic agitation; (3) EDTA 17% (4) EDTA 17% + Passive ultrasonic agitation; (5) EDTA 17% + Saline; (6) 17% EDTA + Passive ultrasonic agitation + Saline. Human blood will be collected from a healthy volunteer and placed inside the root canal using a sterile microbrush. The roots will be kept in a humidifying chamber for 15 minutes until the clot forms. Then, the specimens will be fixed in 2.5% glutaraldehyde, and dehydrated in decreasing series of alcohol. The fibrin characteristics and density of the clots, as well as the morphological characteristics of the platelets and erythrocytes will be observed using a scanning electron microscope. Fibrin densities in all irrigation groups will be assessed using the ImageJ software and analyzed statistically.
