Evaluation of the role of orange juice admnistration on dyslipidemia and hepatic and cardiac alterations induced by doxorubicin in rats

Abstract

Doxorubicin (DOX) is one of most eficcient antineoplastic, widely used as an anti-tumor drug in many chemotherapic treatment regimens. Although it is substantial therapeutic potential, DOX induces several side effects. The genesis of side effects is mainly due to the production of reactive oxygen species. Thus the suplemmentation of orange juice whose is known for it is antioxidant potential is eligible as an alternative to reduce DOX side effects. This study intends to research the role of orange juice suplemmentation in dyslipidemia and hepatic and cardiac changes induced by DOX in rats. This research aims to evaluate the role of the administration of orange juice on dyslipidemia and hepatic and cardiac changes induced by DOX in rats. 32 male Wistar rats will be used, separated into 4 groups: Control (C), DOX (D), suplemmentation with orange juice (OJ) and suplemmentation with orange juice + DOX (DOJ). Animals will pass trough an 4 weeks treatment period in wich it will have water (groups C and D) or orange juice (groups OJ and DJC) to drink ad libitum. At the end of each week the animals will receive an intraperitoneal injection of 4 mg/Kg of DOX (groups DOJ and D) totaling the dose of 16mg/kg, or equivalent dose of saline solution (groups OJ and C). At the end of the protocol, animals will undergo to ecocardiography to the evaluation of cardiac function and morphology. Then, animals will be euthanized to the removal of blood, heart, and liver. Will be measured total cholesterol, HDL, triglycerides and liver injury biomarkers in the blood serum. The protein expression of GLUT-4, PPAR-Y, CPT1 and TNF will be assessed in the organs . The hepatic tissue will be stained with Oil-red to quantify lipidic content. The statistical analysis will be done by ANOVA TWO-WAY with 5% of statical significance. (AU)
