Abstract

In the pharmaceutical field, several variables and processes can affect the properties and characteristics of new formulations. Therefore, the use of powerful algorithms during data analysis allows adoption of proper mathematical models as a strategy to reduce the complexity of the subject and increase our knowledge of the overall system. During this BEPE, ComDim algorithm will be used to evaluate the relative importance of a variable in a data table and extract chemical and physical information on a new drug delivery system (DDS) based on a nanostructured lipid carrier (NLC). Data will be collected from: Raman spectroscopy imaging and small angle X-ray scattering (SAXS) coupled to thermal analyzes - differential scanning calorimetry (DSC) and thermal gravimetric analysis coupled to Fourier transform infrared spectroscopy and mass spectroscopy (TGA-FTIR-MS). Each of these techniques alone provide complementary information which can be easily lost in a simpler analysis. On the other hand, multivariate analysis, including multi-block, is designed to handle the correlation between series of measured variables, revealing the important, underlying (so-called latent) phenomena. The development of new nanopharmaceutical formulations requires information on physical and chemical features of the system. In this sense, the alterations of bulk compounds during the nanoparticles production give insights about materials' behavior and understanding these physicochemical changes is of crucial importance. Here Raman imaging spectroscopy, a relatively new technique, will allow evaluating the spatial distribution of compounds based on their spectra, while SAXS will give information about the nanoparticle organization. On the other hand, thermodynamics, the scientific milestones for investigating the properties of materials, will provide information regarding stability and impurities. It is foreseen that, based on higher comprehension of the processes involved in the DDS development, better medicines to pain treatment will be obtained. (AU)
