Abstract

Recent evidence suggests that intense physical exercise (IPE) can be an important tool for appetite control and possible obesity treatment, since IPE promotes a great acute disturbance in the cytokines (pro and anti-inflammatory) stimulated from muscle tissue, in addition IPE also promotes release of several appetite-modulating neuropeptides in a dose-dependent manner. Both these disturbance in pro and anti-inflammatory system and acute release in appetite-modulating neuropeptides by IPE can result in reduced calorie intake. Also, L-leucine supplementation has the potential to modulate appetite - by activating the mTORC pathway - in addition, to inducing a plasma increase in appetite-modulating neuropeptides, also increases insulin sensitivity. The aim of this study will be to evaluate the acute effect of the association of L-leucine supplementation with sprint interval training (SIT) on appetite control in obese adults. The study will be double blind, randomized, balanced and crossover. Participants will do four experiments: 1) SIT-associated leucine supplementation (SIT + leu), 2) SIT-associated placebo supplementation (SIT + PLA), 3) leucine supplementation without exercise and 4) supplementation of placebo without exercise. Supplementation will consist of three daily doses of 70 mg/kg of leucine or placebo (on the day of training and one day after). The day before, on the day of the SIT and one day after the volunteers will record their food intake. Immediately before the SIT session, immediately, 30 and 60 minutes after, the perception of appetite, blood glucose, GLP-1, PYY, CCK and ghrelin and the immune response (IL1-beta, IL-6, IL-10 and TNF-±) will be evaluated. The data will be analyzed using analysis of variance (ANOVA) of repeated measures (4 treatment groups: SIT + PLA, SIT + leu, SED + Leu + and SED + PLA × points in time), with Bonferroni's post-test, being the significance value of p d0.05.
