Diurnal variation of transcripts and metabolites of sugarcane and energy-cane: an integrated analysis of transcriptomics and metabolomics

Abstract

Energy-cane is a commercial hybrid that has a high fiber content, low sucrose content and greater productivity compared to commercial sugarcane hybrids. In Brazil, some companies have developed commercial varieties of energy-cane, such as CanaBertix® from GranBio S/A. We recently explored the molecular differences between sugar cane and energy-cane through analyzes of sugar metabolism and the biological pathways of primary and secondary metabolism (metabolomics). As a result of the comparison between sugarcane and energy-cane, we identified that sugars (sucrose, glucose, fructose and starch) and some metabolites undergo a phase shift during the daytime variation in leaf (source) and stem tissues (sink), and many biological pathways of energy-cane are more active in different periods of time in relation to sugarcane. These results generated new hypotheses about how the differences in growth, development and productivity of energy-cane in relation to commercial sugarcane hybrids are supported by the dynamics in the production of carbohydrates and metabolites that occurs during the day cycle. However, as the work was based on metabolomics, it was not clear how the regulation of genes related to circadian rhythm and the identified metabolic pathways occurs. In this context, the objective of this project is to analyze the diurnal variation of transcripts and metabolites of the tissues of the leaf and stalk of energy-cane and sugarcane, integrating the data of transcriptomics and metabolomics, to understand why some pathways are more active in energy-cane in different periods of the day cycle compared to sugarcane, and vice versa. For this, we conducted an experiment in which we collected the tissues of the leaf and stalk of sugarcane and energy-cane during the 24-hour day cycle. We will evaluate the expression of genes during the day cycle, in particular, the genes related to the circadian cycle, primary and secondary metabolism and understand how these genes are regulated according to the day cycle, through integrated analyzes of transcriptomic-metabolomics and networks co-expression.
