Abstract

To examine the effect of chronic ischemia associated with vascular disorders on bladder function and the effects of roflumilast treatment, we will investigate morphological and functional changes in the bladder, along with assessments of nitric oxide (NO) and hydrogen sulfide (H2S), and their role in urinary bladder relaxation in rats with internal iliac artery obstruction. Histological techniques will be performed to understand tissue morphology under hypoxic conditions and how roflumilast will help, for functional examinations, cystometry and myographs will be used to measure the functionality of the urinary bladder and iliac artery. In addition, we will use the western blotting, RT-PCR, and immunohistochemical staining of NO and H2S gas-transmitters and of the cyclic nucleotide cAMP/PKA, both in the bladder and in the iliac artery. In this way, we will analyze the effects of roflumilast treatment on the iliac artery and urinary bladder of animals with chronic bladder dysfunction related to ischemia due to iliac artery obstruction.
