Abstract

The endothelium is capable of producing and secreting contracting substances (endothelin-1 and prostanoid-PGH2) and relaxing substances (nitric oxide and prostacyclin) that regulate vascular tone. Although historically catecholamines are strongly associated with neural origin, the group led by Prof. Gilberto De Nucci demonstrated the production of these substances by the vascular endothelium of reptiles and mammals. 6-Nitrodopamine, a new endothelial mediator, formed from the availability of nitric oxide and dopamine, had its effect characterized in vascular preparations (in vitro) of reptiles and mammals. However, investigations regarding its role in systemic and renal hemodynamics, as well as the purification and sequencing of its receptor have not yet been carried out. This investigation will represent an advance in cardiovascular physiopharmacology. Purification and sequencing of the 6-ND receptor in rat atria will be critical for pharmacodynamic studies and future development of agonists/antagonists for 6-ND receptors. The present grant application aims to evaluate the in vivo effect of 6-ND on reptiles (Chelonoidis carbonaria, Trachemys scripta and Tupinambis teguxim), to investigate systemic hemodynamics in rats (telemetry) and to investigate renal effects (perfused kidneys and renal hemodynamics), as well as sequencing and purifying and sequencing 6-ND receptors.
