Analysis of the material/dentin interface after different treatments and the interaction with bioceramic endodontic sealers and repair

Abstract

Bioglass is a category of biomaterials that has biocompatibility, high bioactivity and the ability to bind to living tissue. Biosilicate (LaMaV-UFSCar, São Carlos, SP, Brazil) is a bioglass composition whose particles can be used to improve the bonding capacity with dentin and promote bioactivity. The objective of this study will be to develop dentin treatment protocols prior to the use of bioceramic endodontic sealers. The effect of different treatments on dentin and on the sealer/dentin interface will be evaluated: biosilicate solution at different concentrations, PBS and distilled water as a control. Subproject 1: Bovine dentin blocks will be used for evaluation by means of SEM and X-ray energy dispersion spectroscopy (EDS). After different treatments and contact with bioceramic sealer Bio-C Sealer (Angelus, Brazil) and BioRoot RCS (Septodont, France) the sealer/dentin interface will be evaluated. Dentin/sealer blocks will be scanned in micro-CT (9 µm resolution). The bond strength of the filling material with the dentin will be measured by means of the push-out test and microshear bond strength test. The data obtained will be submitted to the appropriate statistical tests, with a significance level of 5%. Subproject 2: Bovine maxillary incisors will be used to evaluate the root dentin/reparative material interface. After removal of 3mm apical, a retrograde cavity will be made where the surface treatments will be made with biosilicate at the concentration selected in subproject 1. The cavities will be filled with Bio-C Repair (Angelus, Brazil) and Biodentine (Septodont, France). Evaluation of the interface/adhesion of the selected materials will be carried out from surface analyzes in SEM, micro-CT (9 µm resolution), bond strength test (Push-out) and bond strength test to microtensile following experimental protocols similar to those described in Subproject 1.
