Evaluation of Tissue Response with the Use of Platelet-Rich Fibrin Isolated and Associated with Different Stimuli in Wounds Repair in Wistar Rats

Abstract

The integumentary system is the largest organ in the body and its main function is to be a barrier against the surrounding environment. Wounds are defined as an interruption of the continuity of the skin, which can affect one or more layers. Currently, there are several types of materials available on the market to treat these injuries. One of them is the LASER, formed from the emission of electromagnetic radiation, with the objective of promoting tissue regeneration, reducing inflammation, relieving pain, angiogenesis and epithelial proliferation. Another example is an Ovine Forestomach Matrix (OFM), represented here as the Endoform® dermal membrane, considered a decellularized extracellular matrix biomaterial. It acts at all stages of healing to stabilize, correct, create and organize tissue. Finally, we have Platelet-Rich Fibrin (PRF), a biomaterial that has platelets, cytokines, growth factors and leukocytes that act by releasing high concentrations of bioactive structural proteins, acting as a healing promoter of temporal release in angiogenesis and tissue remodeling. The objective is to comparatively evaluate, in a macro and microscopic way, the skin tissue regeneration process in Wistar rats, from the use of PRF alone or associated with other methods. Thus, 75 rats will be used, divided into 3 large groups based on the analysis period (3, 15 and 30 days). In each group, they will be subdivided into 5 other groups, each one with 5 animals, corresponding to the treatment model: control group, group with isolated PRF, group with PRF and laser, group with PRF and OFM and a group with PRF, laser and OFM. A critical defect will be performed on the back of the rats and the treatment will be applied. At the end, samples will be collected for histological evaluation. Data will be evaluated using analysis of variance ANOVA for macroscopic comparison of the size of the wounds. Semi-quantitative non-parametric test (KrussKall-Wallis) and post Mann-Whitney test will also be performed.
