Abstract

The BEPE project aims to complement, with confocal microscopy, the experiments that are being carried out with grapevine leaf rust (Neophysopella tropicalis) in 'Niagara Rosada' grapevine under abiotic stresses. 'Niagara Rosada' plants with water deficit and thermal stress will be inoculated with a suspension of N. tropicalis, adjusted to the concentration of 105 urediniospores mL-1. Leaf samples, collected at 1, 2, 9, 15 and 20 days post-inoculation, will be fixed in 0.15% trichloroacetic acid in 4:1 ethanol/chloroform. The samples will be sent to RWTH Aachen University, where the WGA-AF488 and Propidium Iodide co-staining protocol will be applied and confocal laser scanning microscopy performed. The technique will allow the visualization of the pathogen in the host tissues and the reconstruction of 3D images. These evaluations will be useful to better understand the joint effect of abiotic and biotic stresses on the host-pathogen interaction. The BEPE will be carried out at RWTH Aachen, Germany with the supervision of Professor Dr. Ulrich Schaffrath, a specialist in histological and molecular studies of non-host plants subjected to infection by pathogens, with a broad repertoire in the study of rusts, over a period of 2 months.
