Abstract

In industries in general, lipases play crucial roles in several applications, including the synthesis of biodiesel, modification of fats and oils, production of functional foods, formulation of enzymatic detergents, and in the pharmaceutical industry for the production of specific medicines, in addition to prolonging the shelf life of products, improve the flavor and texture of food, and act in biotechnological cleaning processes. Specific enzymes, such as lipase, require high purity due to their industrial applications. Generally, to obtain a 30% yield and a high purification factor, four to five purification steps are required. This implies a purification process that will certainly have a negative economic impact on the final cost of the product, as the downstream process can represent up to 80% of the total production costs of microbial enzymes. Therefore, many conventional methods are not viable due to the high cost and undesirable impacts. In this sense, a purification protocol that was non-traditional to classical chromatographic processes and at a considerable lower cost was developed. This new purification method involves a single step where the lipase is placed in contact with DEAE-Cellulose for selective binding and subsequent recovery of the pure enzyme. This technology, developed in our research group, was deposited at the National Institute of Industrial Property - (INPI), according to patent application BR 10 2019 006621 0. Therefore, this work aims to improve this purification technology, improving the parameters described in the patent and adding details to the process.
