Abstract

Oxidative stress is characterized by an imbalance between antioxidant factors and free radicals, which can be harmful to the body by causing cellular damage and inducing autophagic and inflammatory pathways. During the postprandial period, a pro-oxidant state is observed due to increased production of reactive oxygen species (ROS) in the mitochondria. During this period, a transient inflammatory response is also observed, characterized by an increase in circulating leukocytes, changes in their gene expression, and an increase in the plasma concentration of inflammatory cytokines. The aim of this study is to characterize oxidative stress during the postprandial period in women. To achieve this, 40 women will be recruited and subjected to a dietary challenge followed by the collection of 10 venous blood samples over 6 hours for the extraction of plasma and leukocytes. Oxidative capacity in plasma will be assessed using DPPH (2,2-diphenyl-1-picrylhydrazyl) and determination of reduced/oxidized glutathione (GSH/GSSG), as well as determination of ferric reducing power, radical oxygen absorption capacity, and nitric oxide (NO) concentration through colorimetric enzyme assays. Gene expression of parameters related to oxidative stress in PBMCs will also be evaluated. Understanding the relationship between oxidative stress and transient inflammation during the postprandial period will deepen the understanding of the mechanism of chronic diseases associated with an imbalance in energy metabolism, as well as identify potential diagnostic markers, aiding in the early detection of chronic diseases.
