Integrative analysis to identify different genes and biological processes modulated in osteoblasts grown on titanium with nanotopography in the presence of osteoclasts

Abstract

Interactions between osteoclasts and osteoblasts are essential for the bone remodeling process and consequently for the osseointegration of titanium (Ti) implants. Several studies have demonstrated the role of the Ti surface with nanotopography (Ti Nano), obtained through conditioning with H2SO4 and H2O2, in the modulation of signaling pathways and molecules involved in the osteogenesis process; nonetheless, several biological processes, signaling pathways and genes modulated during osteogenic differentiation on this surface, in the presence of osteoclasts, remain unknown. In this context, the objective of this study is to perform an integrative analysis, through the use of RNA sequencing (RNA-Seq) data and bioinformatics tools, to identify distinct biological processes, signaling pathways and genes modulated in osteoblasts cultured on Ti Nano in the presence of osteoclasts. For this purpose, data deposited in a functional public repository will be used for differential expression analysis by DESeq2 and enrichment by Gene Ontology (GO). To consider the differential gene expression, we will use Fold Change >1.7 and adjusted p-value <0.05. The development of this project may contribute to the comprehension of different cellular events that mediate bone remodeling and that may contribute to the osseointegration process. (AU)
