Abstract

This project is a research proposed to the FAPESP post doctored program. This project will be development in the Universidade Estadual Paulista Júlio de Mesquita Filho -UNESP, in the Biosciences, Arts and Exact Sciences Institute joined on Chemistry and Environmental Science Department. During doctorate and master period were realized studies about screening, production, purification and characterization of the microbial cyclodextrin glucanotransferase (CGTase) enzyme. In this proposed research, it is intend to continue these studies, investigating this CGTase to industrial application. The doctored studies were realized in the UNESP on Bioscience Institute, campus of the Rio Claro, and in this period two microbial CGTases were purified and characterized. For continuing the doctored studies, it is proposed, in a first step, studies about cloning, expression and sequencing of the CGTase gene as soon as production, purification and characterization of the recombinant CGTase. In a second step, it is intend to realize studies about three-dimensional structure this enzyme using a structural homology and realize study of site-directed mutagenesis, improving interesting properties of industrial application. In a third step, it is intended to produce and characterize the cyclodextrins kind. (AU)
