Abstract

This work aims to optimize the conditions of amplification and revelation of microsatellite markers, as well as to evaluate the genetic diversity of 17 accessions (landraces) of yam (Dioscorea cayenensis Lam.) collected in authoctone agricultural systems of the Vale do Ribeira (SP), in the municipalities of Cananeia, Iguape, Iporanga and Eldorado. For the DNA extraction the method of Elias al. (2004) will be tested, based on the CTAB extraction buffer, with modifications. The conditions for the amplification and revelation of 13 primers will also be tested for the evaluation of 17 accessions of this species. Being a polyploid species, data will be genotyped as presence and absence (+/-) of bands and submitted to a variance analysis (AMOVA) using the Arlequin program (Schneider et al., 2000). Also, a cluster analyses will be conducted, using the coefficient of Jaccard and the UPGMA method (Alfenas, 1998), with the software NTSYS-PC (Rohlf, 1992), and an analysis of principal components, with the software SAS for windows.
