Abstract

The irradiation with laser in low intensity has shown to a therapeutical factor after conventional treatments, reducing the time of healing and acting as anti-inflammatory and analgesic. This irradiation has specific wave lengths that modify the cellular behavior acting in the mitochondria and the calcium channels Osteoblasts are young cells with intense metabolic activity and responsible for the production of organic part of the matrix (collagen type I, glycoproteins and proteoglycans). With respect to the effects of laser in osteoblasts, the studies have shown positive results in the increase in the production of alkaline phosphatase, osteocalcin, bone osteopontina and sialoprotein. It also has a expressive increase in the adhesion and proliferation of the osteoblasts and production of mineralized nodules. The objective of this work will be to evaluate the influence of the phototherapy with laser in low intensity in the viability and enzymatic activity of osteoblasts and if exists influence of this therapy in the expression of RANKL and OPG produced by these cells. For the experiment will be used osteoblasts of lineage MC3T3 ATCC) that will be plaques on plates of 96 wells and in them will be irradiated red laser (660nm) and infrared (780nm) at doses of 3 and 5 J / cm ², beyond a control group without irradiation. For the analysis of the cellular viability it will be made the reduction of MTT and capitation of the Neutral Red; for the analysis of the expression of RANKL and OPG will be made the quantification of transcripts for RT-PCR. The statistical analyses will be made through the statistical test one-way ANOVA complemented for the test of Tukey, test of Kruskal-Wallis complemented for the test of Dunn and for the "t" test, according to the indication (p <0.05) .Key-word:Cellular cuture, photherapy, laser of low intensity, osteoblasts MC3T3.
