Evaluation of bioaccessible and bioavailable fraction of nutrients and contaminants in beef cuts

Abstract

This study describes the determination of total, bioaccessible and bioavailable fractions of Al, Cd, Cr, Co, Cu, Fe, Mo, Pb, Se and Zn contents in beef samples. Liver, muscle and bottom round samples from the same animal were evaluated under three initial conditions: freeze dried, unprocessed (raw / in nature), and cooked (with water and/or fried). All these samples were analyzed by mass spectrometry with inductively coupled plasma after acid mineralization (total content), after sample preparation using an in vitro gastrointestinal digestion method (bioaccessible fraction) and with Caco-2 cells (bioavailable fraction). The contents of several major constituents in different cuts of beefs, such as proteins, lipids, carbohydrates and water, were also measured. Certified reference materials and addition and recovery tests were employed to assess accuracy of the proposed methods, and the recoveries for all elements ranged from 87% to 108%. The total concentration of all elements determined in this study was similar with other reported studies and the measured Cd and Pb concentrations were below the values established by European and Mercosul legislation. The bioaccessibilities values of elements varied with the sample preparation mode (freeze dried / cooked / raw), although the concentrations of all elements were much higher in freeze-dried samples than in all other sample preparations. The bioavailable concentrations for most studied elements were below the limit of quantification, but it was possible to estimate the bioavailable fraction of Al, Fe, Zn and Cu ranging from <LOQ to 500 ìg kg-1 in the different beef samples (AU)
