Development, stability and component study of a deconstructing bacterial consortium of lignocellulose

Abstract

The production of second generation (2G) ethanol obtained from lignocellulosic material to be efficient and economically viable needs to find better options in the process of turning cellulose and hemicellulose into hexoses and pentoses so that they can be fermented. Thus, the objective of this work was to study organisms that make up the bacterial consortium, with the ability to deconstruct lignocellulosic biomass releasing sugars. Cultivation was carried out in BHB (Bushnell Haas Broth) medium with sugarcane bagasse for 5 and 10 days for the 2nd and 20th week of cultivation. Scanning electron microscopy analyzes were performed to evaluate bacteria adherence to the fiber. In addition to evaluating the composition of the residual fibrous fractions of the crop and determining sugars, these analyzes showed that with 5 days to the 2nd week there was greater lignocellulose degradation and sugar release. The bacterial populations present in the consortium were studied by partial sequencing of the 16S rRNA gene, showing that there are genera with the ability to degrade biomass, such as Caulobacter, Pseudoxanthomonas, Chryseobacterium, Sphingomonas, Rhizobium and Dokdonella. Through the studies, it can be seen that the consortium managed to degrade the biomass and release glucose in the culture medium, thus constituting a tool that can be applied in processes of obtaining second generation ethanol.
