Characterization of homologies between bovine BAC probes (Bos taurus) and gray brocket deer (Mazama gouazoubira) chromosomes: a new perspective on intra and interspecific studies of the genus Mazama

Abstract

The Cervidae family presents a marked karyotypic variation as a reflection of its high rate of chromosome evolution. The Neotropical deer from the genus Mazama stands out for its expressive chromosomal polymorphism. Although the species of the genus differ in the values of their diploid number (2n), they are closely related and similar morphologically. Due to this, the group exhibits taxonomic uncertainties. The chromosomal evolution of the Mazama species from the hypothetical ancestral karyotype, retained by the gray brocket Mazama gouazoubira (2n = 70), is poorly understood and lacks molecular cytogenetic analysis. Then, the present study revealed chromosomal homologies between bovine (Bos taurus, 2n = 60) and M. gouazoubira using a combination of bacterial artificial chromosome (BAC) clones derived from cattle. By fluorescent in situ hybridization, we constructed a cytogenetic map with 106 new markers to the 34 gray brocket autosomes and the X chromosome. Our results showed that bovine BAC probes proved to be satisfactory for the establishment of chromosomal markers in deer. The evolutionary differences between the bovine and gray brocket deer chromosomes include five fissions and one tandem fusion in the cattle. Each group of BAC probes derived from bovine chromosomes pairs 1, 2, 5, 6, 8, and 9 presented hybridization signals on two different chromosomes, while pairs 28 and 26 are fused in tandem on a single acrocentric chromosome in M. gouazoubira. For the other chromosomes, the BAC probes derived from the same bovine autosome showed signals in a unique chromosome pair in M. gouazoubira. Moreover, we detected that the homologous to the autosome pair 1 and X chromosome are intrachromosomally rearranged in M. gouazoubira. Therefore, it is expected that future studies using the produced markers result in great taxonomic and reproductive knowledge for brocket deer. (AU)
